三种蓑藓属(Macromitrium)植物DNA C-值测定-兼论苔藓DNA-C值研究的生物学意义

摘要:
DNA C-值是指一个物种没有复制的配子的核DNA含量,是一个重要的生物学性状。关于植物DNA C-值的研究已经为系统发育学、分类学、生态学、生物地理学等提供了重要信息,但是工作主要限于被子植物。苔藓植物是一类特殊的高等植物,关于苔藓植物DNA C-值的研究才刚刚起步。本文分别以小立碗藓(Physcomitrella patens)和日本晴(Oryza sativa spp. japonica)为标样,用流式细胞术分别测定了中华蓑藓(Macromirium cavaleriei)、钝叶蓑藓(M. japonicum)和缺齿蓑藓(M. gymnostomum)三种藓类植物的DNA C-值,由此建立了应用流式细胞术测定蓑藓属植物DNA C-值的方法,讨论了影响藓类植物DNA C-值测定结果的因素。论文最后讨论了苔藓植物DNA C-值变异式样、系统发育和分类学意义,以及今后有关藓类植物DNA C-值研究中值得关注的几个方向。

Applying Flow Cytometry to determine DNA C-value of three Macromitrium species, with comets on biological significances of Bryophyte DNA C-Values

Abstract:
The DNA amount in the unreplicated gametic nucleus of an organism is referred to as its C-value, which is an important biological trait. Research on plant DNA C-values has provided important information for phylogenetic, taxonomic, ecology, biogeography, etc., but the work has been conducted mainly within taxa of angiosperms. Bryophytes are special higher plant taxa. Few research on DNA C-value of bryophytes has been carried out. In this paper, the DNA C-values of three Macromitrium species (including M. cavaleriei, M. japonicum and M. gymnostomum) were determined by flow cytometry by using Physcomitrella patens and Oryza sativa spp. japonica as a standard, respectively. Thus the method of measuring DNA C-values of Macromitrium species by flow cytometry was established. The factors influencing the measuring result were also discussed. Finnally, variaiton patterns of bryophytes DNA C-value, their phylogenetic and taxonomic significance, and some interesting topics about DNA C-value research were remarked.
